Founded in April 1974 at Nawabshah as Peoples Medical College, the premier institution for women's medical education in Sindh under the vision of Prime Minister Zulfikar Ali Bhutto, initially housed temporarily in a District Council High School building. It was upgraded to Peoples University of Medical & Health Sciences for Women by a Government of Sindh act dated 21 December 2009, with the Governor of Sindh's assent on 10 November 2010.

The university is located in Shaheed Benazirabad (formerly Nawabshah), a city in Sindh renamed in 2009 in honour of Benazir Bhutto after her assassination in nearby Rawalpindi, and a longstanding agricultural and administrative centre of the province.
