Founded on 10 December 1945 when Sir Hugh Dow, Governor of Sindh, laid its foundation stone on the site of the old NJV School, converting the earlier Hyderabad medical school (started by Dr. Homestead in 1881); on 31 December 1945, 45 students including one Muslim woman transferred from Hyderabad to Karachi. It was affiliated with the University of Bombay until 1947, then the University of Sindh, and later the University of Karachi, before becoming a founding constituent college of the newly established Dow University of Health Sciences in 2003.
