Founded in the second half of 2008 under Rector Vice Admiral Farooq Rashid, with its first batch of MBBS students beginning that year; it was formally inaugurated by the Chief of Naval Staff, Admiral Noman Bashir, on 29 January 2009. The first MBBS batch graduated in December 2013.

The college is located in DHA Phase 2, Karachi Cantonment, an area developed under the Pakistan Navy and Defence Housing Authority as part of the city's expanding southern coastal districts.
